- HNPP, 0.5 ml HNPP (10 mg/ml; 23.8 mM) in dimethylformamide (DMF)
- Fast Red TR, 100 mg Fast Red TR, powder
In combination with Fast Red, HNPP is used for fluorescent detection of a nonradioactively labeled nucleic acid probe in in situ hybridization. HNPP/Fast Red gives a bright, long-lasting red or orange signal withstanding long exposures to UV light. HNPP alone can also be used for membrane blotting applications in which it gives a precipitable distinct signal. It is especially suited for highly sensitive detection of nonradioactively labeled nucleic acids in
- Fluorescence in situ hybridizations (FISH)
- Southern blot
- Northern blot
- Colony or plaque hybridizations
- Nonradioactive DNA sequencing blots.
The sensitivity of detection of nonradioactive Southern blots is equivalent to NBT/BCIP and CSPD, after a 2-hour to overnight incubation.
