Compared within AMV reverse transcriptase, M-MuLV reverse transcriptase lacks endonuclease activity and has much lower RNase H activity. Full-length copies of large mRNA are obtained.

M-MuLV reverse transcriptase is used for in vitro synthesis of cDNA transcripts of specific RNA sequences; for the preparation of cDNA libraries; or for synthesis of first-strand cDNA for use in subsequent amplification reactions (RT-PCR). M-MuLV reverse transcriptase is used in dideoxy-sequencing reactions in place of Klenow enzyme. Reverse transcriptase will often synthesize through GC-rich regions where Klenow enzyme is hindered.
